  已经开发了用于制备对映体纯的（S）-和（R）-缬氨酸叔丁酯的大实验室规模（约40g）的方法。该方法包括三个步骤：制备N -TFA-缬氨酸，在硫酸存在下使用2-甲基丙烯在二恶烷中制备缬氨酸叔丁酯，以及分离目标化合物作为乙酸酯衍生物。相对于起始缬氨酸，总产率高达70％，ee大于98％（通过HPLC）。

  The dynamic kinetic resolution in the synthesis of alkyl N-acetylphenylalanyl (S)-valinates was studied by the mixed anhydride method. The structure of the ester group of the amino component appeared to exert no substantial effect on the diastereoselectivity of the reaction.

  Reaction of N-2-protected 2-amino-6-chloropurine with tert-butyl (S)-phenylalaninate, (R)- and (S)-valinates followed by deprotection affords 2-aminopurines bearing at 6-position the corresponding amino acid moieties, whose chiral centre is partially racemized.
