This invention relates to a method for identifying maxi-K channel blockers comprising using HEK-293 cells that have been stably transfected with both alpha and beta 1 subunits of the maxi-K channel and incubated with potassium channel blockers that selectively eliminate the endogenous conductances of the HEK-293 cells. Maxi-K channels are important for controlling a number of physiological processes. They play a role in the regulation of aqueous humor dynamics in the eye. Maxi-K channel blockers are known to reduce intraocular pressure.

Ce procédé consiste à utiliser des cellules HEK-293 ayant subi une transfection stable par des sous-unités alpha et bêta-1 du canal maxi-X et une incubation avec des inhibiteurs de canaux potassiques qui éliminent sélectivement les conductances endogènes des cellules HEK-293. Les canaux maxi-K ont une fonction importante pour le contrôle d'un certain nombre de processus physiologiques. Ils interviennent dans la régulation de la dynamique propre à l'humeur aqueuse de l'oeil. On sait par ailleurs que les inhibiteurs de canaux maxi-K réduisent la pression intra-oculaire.
