    The spirolactam ring-opening process of rhodamine derivative is one of the most useful mechanisms for controlling fluorescence properties. However, the open/closed equilibrium reaction of rhodamine spirolactam has not been well characterized. Therefore, we examined the relationship between the spirolactam ring-opening process of rhodamine derivatives and the structure of the xanthene moiety. Based on the results of this investigation, we selected a candidate xanthene moiety for a Zn2+ sensor, and successfully developed a new fluorescence probe for Zn2+. (C) 2010 Elsevier Ltd.     Provided is a compound represented by formula I, wherein R 9 is a C1-C22 hydrocarbyl group, or a C2-C3 ester-substituted C1-C3 alkyl group, R 5 , R 6 , R 7 , and R 8 are alkyl groups or H, and R 1 , R 2 , R 3 , and R 4 are H or a lower hydrocarbyl group. Or, R 9 is a C2-C22 hydrocarbyl group, or a C2-C3 ester-substituted C1-C3 alkyl group, and R 5 is connected to R 1 , R 6 is connected to R 2 , R 7 is connected to R 3 , R 8 is connected to R 4 to form six-membered rings. The present compound is temperature-sensitive and can enter into cells. An intracellular temperature distribution image having high spatial and temporal resolution can thus be obtained. The present compound can also perform distribution calibration on a temperature-sensitive fluorescent compound. Also provided is a method for measuring temperature distribution within a living cell, and a corresponding detection kit. The method satisfies the requirements of small size measurement and rapid measurement, thereby achieving high resolution in terms of space and time.

    Protein profiling probes are important tools for studying the composition of the proteome and as such have contributed greatly to the understanding of various complex biological processes in higher organisms. For this purpose the application of fluorescently labeled activity or affinity probes is highly desirable. Especially for in vivodetection of low abundant target proteins, otherwise difficult to analyse by standard blotting techniques, fluorescently labeled profiling probes are of high value. Here, a one-pot protocol for the synthesis of activated fluorescent labels (i.e.azide, alkynyl or NHS), based on the Ugi-4-component reaction (Ugi-4CR), is presented. As a result of the peptoidic structure formed, the fluorescent properties of the products are pH insensitive. Moreover, the applicability of these probes, as exemplified by the labeling of model protein BSA, will be discussed.

    本发明公开一种含有谷氨酸结构的罗丹明pH荧光探针及其制备方法和应用,所述荧光探针为G‑pH荧光探针,G‑pH荧光探针的结构通式如(Ⅰ)所示。该G‑pH荧光探针在pH4.46‑9.0范围内呈现出较宽的pH响应,并且该探针的pKa为7.36,处于合理的生理pH区间,此外该探针还具有较好的化学、光稳定性,较好的溶解性和生物兼容性,不受金属离子的干扰。显微成像实验表明这类探针具有较好的细胞通透性,对细胞和生物体无毒副作用。
