    A method which comprises using a specified dye taken up by erythrocytic nucleated cells so that their nuclei are stained and can be readily differentiated from other cells by measurement with a flow cytometer is disclosed. The method for differentiating erythroblasts from other cells relies basically upon flow cytometry by two-step staining that uses a first fluid that is an acidic hypotensive fluorescent dye solution and a second fluid that is a solution that changes the osmorality and pH of the first fluid. The first fluid further contains a fluorescent dye that diffuses into erythroblasts to specifically stain their nuclei, and a group of erythroblasts is separated from other cell groups on the two-dimensional plot constructed with the flow cytometer, whereby the results of erythroblast differentiation are computed.

    A method for leukocyte classification which relies basically upon flow cytometry that uses a first fluid that is hypotonic acidic fluorescent dye solution and a second fluid that is a solution that changes the osmolarity and pH of the first fluid is disclosed. In accordance with the method, a fluorescent dye that specifically stains the nuclei of damaged leukocyte cells is added to the first fluid and the resulting fluid is used in measurements. The first fluid to be used in the method incorporates not only the first and second dyes but also the third dye and this insures that even leukocytes with damaged cell membrane can be distributed as entities distinct from other leukocyte types on a two-dimensional plot. And, hence, high precision of leukocyte classification is assured even in the case of assaying blood that contains damaged cells as a result of prolonged standing at room temperature after sampling.

    A process is provided for separating individual microorganisms either from a support such as soil particles to which they are adhered or from a flock in which the microorganisms are adhered together to form groups. The process involves enzymatically decomposing the natural polymeric adhesive which holds the microorganism to the support or to one another. After the individual microoganisms have been separated, they can be counted, or the proportion of one of the microorganisms within the microorganism population can be determined, or nucleic acids can be separated from the microorganisms. The enzyme may decompose polysaccharides, proteins or pectins and mixtures of enzymes may be used. Suitable enzymes for bringing about the separation of the individual organisms comprise cellulase, hemicellulase, glucuronidase, amylase, protease and pectinase.
