    The present invention relates to a process for preparing haptens for immunoassay of phosphorothioate pesticides, which comprises the steps of reacting O-methyl(ethyl) dichlorothiophosphate with a phenolic compound to obtain O-methyl(ethyl) O-aryl chlorothiophosphate, and reacting the O-(methyl)ethyl O-aryl chlorothiophosphate thus obtained with aminocarboxylic acid to give desired haptens. In accordance with the present invention, haptens having a structure of O-methyl(ethyl) O-aryl, N-(carboxyalkyl)phosphoramidothioate or O-methyl(ethyl) O-aryl N-alkyl-N-(carboxyalkyl)phosphoramidothioate can be simply prepared with a high yield by employing two-step processes in a cost-efficient manner.

  • The Simultaneous Determination of Chlorpyrifos–Ethyl and –Methyl with a New Format of Fluorescence-Based Immunochromatographic Assay

    The improper and excessive use in agriculture of chlorpyrifos–methyl (CPSM) and chlorpyrifos–ethyl (CPSE) may affect the health of human beings. Herein, a fluorescence-based immunochromatographic assay (FICA) was developed for the simultaneous determination of CPSM and CPSE. A monoclonal antibody (mAb) with equal recognition of CPSM and CPSE was generated by the careful designing of haptens and screening of hybridoma cells. Instead of labeling fluorescence with mAb, the probe was labeled with goat-anti-mouse IgG (GAM-IgG) and pre-incubated with mAb in the sample. The complex could compete with CPS by coating antigen in the test line. The new format of FICA used goat-anti-rabbit IgG (GAR-IgG) conjugated with rabbit IgG labeled with fluorescence microspheres as an independent quality control line (C line). The novel strategy significantly reduced nonspecific reactions and increased assay sensitivity. Under the optimal conditions, the proposed FICA showed a linear range of 0.015–64 mg/L and limit of detection (LOD) of 0.015 mg/L for both CPSE and CPSM. The average recoveries of CPS from spiked food samples by FICA were 82.0–110.0%. The accuracy was similar to the gas chromatography–tandem mass spectrometry (GC-MS/MS) results. The developed FICA was an ideal on-site tool for rapid screening of CPS residues in foods.

  • Development of Enzyme-Linked Immunosorbent Assays for the Insecticide Chlorpyrifos. 1. Monoclonal Antibody Production and Immunoassay Design

    The present work describes the production and characterization of monoclonal antibodies (MAbs) to the insecticide chlorpyrifos and their incorporation into several ELISA configurations. With this aim, a collection of chlorpyrifos haptens was synthesized by introducing appropriate spacers in opposite sites of the analyte molecular structure. From mice immunized with protein conjugates of these haptens, several hybridomas secreting MAbs with the ability to sensitively bind the analyte were obtained. IMAbs showing the highest affinity to chlorpyrifos in homologous assays (I-50 values in the 20-220 nM range) were selected. Hapten heterology involving modifications of the moiety closer to the attachment site provided the highest improvement in sensitivity. MAbs displayed striking differences in their cross-reactivity pattern with structurally related compounds. One MAb (I-50 around 10 nM) was incorporated into other ELISA formats. No remarkable changes of assay characteristics, other than immunoreagent consumption and immunoassay procedure, were found. These ELISAs are potentially very valuable analytical tools for the rapid and sensitive determination of this insecticide.

    Highly sensitive enzyme-linked immunoassays for chlorpyrifos, one of the most applied insecticides, are presented. Several haptens were synthesized for immunoreagent production and ELISA development. The best immunoassays obtained are based on an indirect coated-plate immunoassay format. Two assays were optimized; one shows a limit of detection of 0.3 ng L-1, an /(50) of 271 ng L-1, and a dynamic range between 4 and 16 474 ng L-1. The other one has a limit of detection of 0.07 ng L-1, an 150 of 7 ng L-1, and a dynamic range between 0.4 and 302 ng L-1. The assays were used to quantify chlorpyrifos in olive oil using a simple and rapid sample extraction procedure. The good recoveries achieved in both assays (109% mean value) and the agreement with values given by the GC reference method (110% mean value) indicate their potential for either screening or laboratory quantification.
