Synthesis of Novel Tryptamine and Azepinoindole Derivatives

A novel one-pot synthesis of azepinoindoles (9) via reaction of tryptamines (4) with formaldehyde is described. The scope and generality of this new procedure and the preparation of the starting compounds are also discussed.

Valuable new synthetic intermediates, 7-chloromethyl-1H-indole-2-carboxylates (3a-d), were prepared by the facile elimination of sulfur dioxide under the influence of thionyl chloride from 2-ethoxycarbonyl-1H-indole-7-methanesulfonic acids (1a-d), easily accessible by Fischer-type indolisation. The 7-chloromethylindoles easily underwent methanolysis and aminolysis.

Galat reactions between aldehydes and substituted malonic acids half oxyester were found to be efficiently catalyzed by morpholine in refluxing toluene. This transformation allows the stereoselective synthesis of diverse alpha,beta-disubstituted acrylates in moderate to good yields. This method constitutes an attractive alternative to existing methods in terms of scope and eco-compatibility.
