    Lipophilic peptide nucleic acids containing a 1,3-diyne function: synthesis, characterization and production of derived polydiacetylene liposomes

    Adenine-, cytosine- and thymine-containing peptide nucleic acid (PNA) monomers have been synthesized in which either diacetylenic or stearoyl moieties are attached to the N-or C-terminus; the diacetylenic group is embedded within a long hydrocarbon chain. A range of analogous lipophilic functionalized PNA oligomers have been prepared using either solid phase synthesis or a post-synthetic solution phase procedure following cleavage of the PNA oligomer from the solid Support. Selected functionalized PNA monomers and oligomers have been incorporated into liposomal polydiacetylenes and characterized by UV-vis absorption spectroscopy. Preliminary investigations show that blue PDA-liposomes containing thymine-based PNAs can be formed and that production of liposomes with other PNA systems are viable. (c) 2005 Elsevier Ltd.   • Synthesis of Alkyne-Terminated PCDA Linker for Applying Click Chemistry on PDA Layers

    An alkyne-terminated PCDA (10,12-pentacosadiynoic acid) linker, which can undergo a simple click reaction with azide compounds, was synthesized for the purpose of immobilizing various kinds of biomolecular surface ligands on PDA (polydiacetylene) layers. In order to confirm the ability of the linker compound to anchor azide-modified surface ligands, we carried out a model ­reaction with benzyl azide under typical click-reaction conditions. We were able to confirm that the model reaction proceeded smoothly by observing the color change of PDA, including its thermochromic reversibility.

    A microarray includes a solid substrate having a surface, the surface having a plurality of binding spots and a plurality of reaction moieties bound to the binding spots. A reaction moiety includes a plurality of polyacetylene monomers, the polyacetylene monomers having a first coupling region and a second coupling region, the first coupling region having a first functional group operable to bind to the binding spot and the second coupling region comprising a second functional group operable to bind to an accessory molecule; and an accessory molecule having a binding region and an analyte reaction region, the analyte reaction region operable to selectively bind to the target analyte, and the binding region operable to bind to the second coupling region of the polyacetylene monomer. Upon binding a target analyte with the reaction moiety, a color change from the polyacetylene monomer occurs and the reaction moiety produces fluorescence.
