    Novel Class of Benzoic Acid Ester Derivatives as Potent PDE4 Inhibitors for Inhaled Administration in the Treatment of Respiratory Diseases
    摘要:
    The first steps in the selection process of a new anti-inflammatory drug for the inhaled treatment of asthma and chronic obstructive pulmonary disease are herein described. A series of novel ester derivatives of 1-(3-(cyclopropylmethoxy)-4-(difluoromethoxy)phenyl)-2-(3,5-di-chloropyriclin-4-yl) ethanol have been synthesized and evaluated for inhibitory activity toward cAMP-specific phosphodiesterase-4 (PDE4). In particular, esters of variously substituted benzoic acids were extensively explored, and structural modification of the alcoholic and benzoic moieties were performed to maximize the inhibitory potency. Several compounds with high activity in cell-free and cell-based assays were obtained. Through the evaluation of opportune in vitro ADME properties, a potential candidate suitable for inhaled administration in respiratory diseases was identified and tested in an in vivo model of pulmonary inflammation, proving its efficacy.

    Disclosed is a copolymer capable of forming an optically anisotropic melt comprising recurring structural units of Formulas I, II, and optionally III wherein each R is independently a chemically inert substituent; and each R¹ is independently benzoil or phenoxy. The present invention also relates to a process for preparing the copolymer. The copolymer of the present invention usually have a melting point below 350°C and can be readily melt extruded into fibers, films or the like.

  • Aiding chronic written language expression difficulties: A case study

    This paper describes some progress made in helping a young aphasic man (MD) to overcome what had appeared to be intractable written language expression difficulties, which continued to be significant for him. Traditional therapy methods based on cognitive neuropsychological assessment had failed to help MD to generalize improvement in his spelling particularly, but not exclusively, of longer and irregular words. Both a splint which allowed him to use his dominant hand for writing directly onto a computer screen and a simple word-processing programme with synthesized auditory feedback and lexical and grammatical prediction (Write:OutLoud(R) and Co:Writer(R)) enabled him to produce more normal written output (increased quantity and more normal quality). The discussion also focuses on how effectiveness of language therapy can be measured in this man with chronic aphasia.

    A series of 1,2,3,4-tetrahydroisoquinoline-3-carboxylic acid diamides that increase chloride transport in cells expressing mutant cystic fibrosis transmembrane conductance regulator (CFTR) protein has been identified from our compound library. Analoging efforts and the resulting structure-activity relationships uncovered are detailed. Compound potency was improved over 30-fold from the original lead, yielding several analogs with EC50 values below 10 nM in our cellular chloride transport assay. (c) 2005 Elsevier Ltd.
