Enantioselective conjugate addition of azlactones to ethylene sulfonyl fluoride has been achieved via the cooperative catalysis with (DHQD)2PHAL and a hydrogen-bond donor (HBD). This approach furnishes a facile access to a range of structurally diverse azlactone sulfonyl fluoride derivatives with good to excellent yields and enantioselectivities. pyrrolidine-catalyzed three-component reaction of azlactone, N,O-acetal and alcohol for the synthesis of α,β-diamino ester was reported. The N,O-acetal served as the imine equivalent to occur Mannich reaction with azlactone, and the in situ generated α-functionalized azlactone subsequently underwent a ring-opening process in the presence of alcohol. A series of α,β-diamino esters were obtained in 36–99% yields
The invention concerns novel amide derivatives of the formula:
wherein R1, R2, R3, R4, R5, R6, R10, R11 and R16 are various substituents defined hereinafter, Y is carbonyl, thiocarbonyl, sulphonyl or amido, X is carbonyl, hydroxymethylene, thiocarbonyl or oximinomethylene and Q is carbonyl or methylene, or a salt thereof, which are inhibitors of angiotensin converting enzyme and may be used for example, in the treatment of hypertension. The invention also provides pharmaceutical compositions of and processes for the manufacture of the novel amide derivatives.

AbstractWith the combined use of silver carbonate, diaryliodonium bromides were successfully utilized as aryl source to realize the highly efficient arylation of azlactones under mild conditions. The reaction worked well with various alkyl‐ and aryl‐substituted azlactones, providing a range of α‐tetrasubstituted amino acid derivatives in moderate to high yields.magnified image
