Exploration of Biaryl Carboxylic Acids as Proton Shuttles for the Selective Functionalization of Indole C–H Bonds
摘要:
A survey of diversely substituted 2-arylbenzoic acids were synthesized and tested for use as proton shuttle in the direct arylation of indoles with bromobenzenes. It was found that 3-ethoxy-2-phenylbenzoic acid gives superior yield and selectivity for this class of substrates.

We describe a powerful, broadly applicable cross-coupling protocol that enables carbon–carbon bondformation at highly sterically hindered carbon centers (both sp2 and sp3) by employing organocopper reagents under palladium catalysis. Experimental studies and theoretical calculations indicated that the key to the unique reactivity of copper is the relatively low activation energy of the compact transmetalation

unsymmetrical, biarylcouplingreaction for hindered systems based on Suzuki methodology has been developed. A description of the optimisation of this process leading to the recommendation of aqueous thallium hydroxide as base, in dimethylacetamide, with hindered boronic acids and aryl halides to achieve good to moderate yields of hindered biaryls is presented, along with an interpretation of the reaction conditons

Using a reverse order of addition of reagents, PdCl2 and Pd(OAc)(2) are efficient catalysts for the Suzuki-Miyaura reactions in water. The ligandless and mild conditions, the high stability of the catalytic system, short reaction time and good to excellent yields are important features of this protocol.
