We describe a new method to produce histone H2B by semisynthesis with an engineeredsortase transpeptidase. DNA–histone cross-links (DPCs) have been synthesized efficiently via oxime ligation. These DPC lesions block DNA replication but the replication block can be removed via proteolytic processing.
