(5R)-6(Z)-[(1-Methyl-1,2,3-triazol-4-yl)methylene]penem-3-carboxylic acid monohydrate (BRL 42715) is a potent beta-lactamase inhibitor. We report a study on the hydrolysis of the four-membered lactam ring of this compound mediated either by Tris buffer or by Tem-2 beta-lactamase. In both studies free-zone capillary electrophoresis proved to be ideal in the identification of the products of reaction, the interpretation of the mechanism of hydrolysis, and the determination of kinetic parameters and stoichiometry.
