  (EN) A class of 1,4-disubstituted piperazine derivatives, further substituted on one of the carbon atoms of the piperazine ring, are selective agonists of 5-HT1-like receptors, being potent agonists of the human 5-HT1D$g(a) receptor subtype whilst possessing at least a 10-fold selective affinity for the 5-HT1D$g(a) receptor subtype relative to the 5-HT1D$g(b) subtype; they are therefore useful in the treatment and/or prevention of clinical conditions, in particular migraine and associated disorders, for which a subtype-selective agonist of 5-HT1D receptors in indicated, whilst eliciting fewer side-effects, notably adverse cardiovascular events, than those associated with non-subtype-selective 5-HT1D receptor agonists.(FR) Cette invention concerne une classe de dérivés de pipérazine 1,4-disubstitués, lesquels sont également substitués sur l'un des atomes de carbone de l'anneau de pipérazine. Ces dérivés sont des agonistes sélecteurs des récepteurs du type 5-HT1, ainsi que des agonistes puissants du sous-type de récepteurs 5-HT1D$g(a) chez l'homme, tout en ayant une affinité élective d'ordre 10 au moins envers le sous-type de récepteurs 5-HT1D$g(a) par rapport au sous-type 5-HT1D$g(b). Ces dérivés sont donc utiles dans le traitement et/ou la prévention d'états cliniques, en particulier des migraines et de troubles associés à ces dernières, pour lesquelles un agoniste sélecteur de sous-type de récepteurs 5-HT1D est indiqué. En outre, ils engendrent moins d'effets secondaires, notamment des incidents cardio-vasculaires, que ceux occasionnés par des agonistes non sélecteurs de sous-type de récepteurs 5-HT1D.
