    Analogs of platelet activating factor. 7. Bis-aryl amide and bis-aryl urea receptor antagonists of PAF

    A series of bis-aryl amide (13-57 and 66-81) and bis-aryl urea (58 and 85) antagonists of platelet-activating factor (PAF) was prepared that contain, separating the two aromatic rings, linear amide linkages of the form -(CH2)nCONH- (n = 0-2), -OCH2CONH-, and -(CH2)nNHCO- (n = 0-1), branched amide linkages of the form -(CH2)nN(COR)- (n = 1-3, R = CH3 or n-C3H7), and -N(COCH3)CH2-, and urea linkages of the form -NHCONH- and -CH2N(CONHCH3)-. These compounds were examined for their ability to inhibit PAF-induced platelet aggregation of rabbit platelets. These in vitro data were compared to similar data obtained for a number of known PAF antagonists. The compounds were evaluated in vivo, in the mouse, for their ability to prevent death induced by a lethal challenge of PAF. The relationships between the biological activity and the nature, lipophilicity, and position of substituents of the aromatic rings were studied. Best activity was observed for compounds having linkages of the type -CH2CONH-, -CH2N(COR)-, and -CH2NHCO-. Many of these compounds inhibit PAF-induced platelet aggregation with IC50's under 1 muM.

  • Discotic metallomesogens: mesophase crossover of columnar rectangular to hexagonal arrangements in bis(hydrazinato)nickel(II) complexes

    Three series ofN-(3,4-dialkoxybenzylidene)-N′-(3′,4′,5′-trialkoxybenzoyl)hydrazine and their nickel(ii) complexes, bis[N-(3,4-dialkoxybenzylidene)-N′-(3′,4′,5′-trialkoxybenzoyl)hydrazinato]nickel(ii), were prepared and characterized. The mesomorphic properties of these disc-like compounds were also studied in terms of liquid crystallinity. The orange nickel(ii) complexes with ten soft alkoxy side chains exhibited columnar disordered mesophases, which were characterized based on DSC analysis and optical polarized microscopy. Nickel complexes with shorter side chains (n=5–8) displayed a monotropic columnar rectangular disordered (Colrd) phase; however, complexes with longer side chains showed monotropic (n=10) and enantiotropic (n=12, 14) columnar hexagonal disordered (Colhd) phases. Similar nickel complexes (n=14) with eight side chains exhibited a columnar hexagonal phase, whereas complexes (n=14) with six side chains formed crystalline phases. The structures of these columnar disordered phases were confirmed by X-ray powder diffraction. XRD data indicated that mesophase crossover from columnar rectangular (Colr) to hexagonal arrangements (Colh) was observed with increasing side chain lengths.

    Several new enaminoketone mesogens were synthesized and coordinated to various metal salts to yield a variety of novel rodlike metal-containing liquid crystals. The mesophases were characterized by polarized optical microscopy (POM), differential scanning calorimetry (DSC) and small-angle X-ray scattering (SAXS). Whereas the free ligands display SmA and nematic mesophases, the NiII and PdII complexes give rise to SmC and nematic phases, and the paramagnetic CuII mesogens to a narrow temperature-range nematic mesophase only. The magnetic behaviour of the latter was studied by means of EPR and magnetic susceptibility (SQUID) measurements.

    assess in solution the presence of the complexes and their tetrameric composition. All the complexes display mesogenic ability, and their mesomorphic organization has been studied by X-ray diffraction and CD spectroscopy. Results allow us to propose a helical columnar model for the mesophase originated from a propeller-like conformation of the supramolecular complexes.

    The merger of ionic liquid and liquid crystal fields, obtained by using the imidazolium ring as a common element, has allowed us to tailor a new set of materials which associate specific functionalities. These functionalities are consequences of the original properties of the component, ionic liquids, liquid crystals and their association in a single compound. The study of this interesting association led us to elaborate environment-flexible cationic architectures from which mesomorphic properties emerge. Moreover, we have also explored the influence of different anions on the mesomorphic properties.
