The first synthesis of justicidone (4-(1′,3′-Benzodioxol-5′-yl)-6-methoxynaphtho[2,3-c]furan-1,5,8(3H)-trione) was carried out from piperonal, as a starting compound, through a lineal process using well known reactions.

A new class of lignan analogues has been synthesized by means of tandem conjugateaddition‐alkylation of butenolides. Furan and thiophene derivatives have been obtained by this method. This type of compounds has been called heterolignanolides and the generic name heterolignans is proposed for this class of lignan analogues.

Cyclization with CF3CO2H of the dibenzyllactone compounds 3 and 4 afforded the aryltetralin derivatives 6a and 6b, epimers at the carbon α to the lactonic carbonyl group, in which the aryl substituent group is in the α-axial position. 1H NMR data assignments of the compounds obtained are also given.

A new class of heteroaromatic analogs of lignans, called heterolignanolides, have been tested against several tumor cell lines. These compounds carry diverse heterocyclic rings, instead of the trimethoxyphenyl ring found in the natural lignans yatein and podorhizol. They have moderate antineoplastic activity (P‐388, A‐549, HT‐29) compared with that of yatein. None of the tested compounds has discernible
