This invention relates to compounds and methods for the inhibition of HDAC enzymatic activity. More particularly, the invention provides for compounds of formula (I), (I) and N-oxides, hydrates, solvates, pharmaceutically acceptable salts, prodrugs and complexes thereof, and racemic and scalemic mixtures, diastereomers and enantiomers thereof, wherein L, M, n, R, W, X and Y are as defined in the specification.

Double Michael addition of dithiols such as 1,3-propanedithiol and 1,2-ethanedithiol to α,β-acetylenic carbonyl compounds in the presence of molecular sieve and dimethyl sulfoxide proceeds very smoothly to afford the corresponding β-keto 1,3-dithianes and β-keto 1,3-dithiolanes, respectively, in good to high yields.
Highly regio- and stereoselective hydrosilylation applicable to a broad range of electron-deficient alkynes has been established using palladium catalysis. Conditions are reported for the facile, high-yielding coupling of acyl chlorides with terminalalkynes in a reaction involving palladium and copper iodide; the reaction is tolerant of a wide variety of acyl chlorides and terminalalkynes and provides a convenient one-pot route to acetylenic ketones.
