  The Cu2O-catalyzed reaction of α,β-unsaturated isonitrile with active methylene compound is described. Vinyl isocyanide (VIC) and propenyl isocyanide (PPIC) were employed as the isocyanide components. In addition, allyl isocyanide (AIC), which has been known to be rearranged readily to PPIC by Cu2O, was also used as a precursor of PPIC. Five types of products were isolated and identified.   ABSTRACT A method has been worked out to synthetize hitherto unknown 5-(2-chloroalkyl) Meldrum's acids 4 starting from easily accessible 2-chlorocarboxylic acid chlorides 1 in a two-step reaction.   A series of 1-sulfonylaminocyclopropanecarboxylates was synthesized as ADAMTS-5 (Aggrecanase-2) inhibitors. After an intensive investigation of the central cyclopropane core including its absolute stereochemistry and substituents, we found compound 22 with an Agg-2 IC(50) = 7.4 nM, the most potent ADAMTS-5 inhibitor reported so far.
