    In one aspect, this invention relates to production of useful fermentation end-products from biomass through simultaneous hydrolysis and fermentation by a microorganism, such as Clostridium phytofermentans . The invention also relates to the development of a process for efficient pretreatment and conversion of lignocellulosic biomass to end-products with high conversion efficiency (yield). In another aspect, methods for producing a fermentation end-product by fermenting hexose (C6) and pentose (C5) sugars with a microorganism, such as Clostridium phytofermentans are disclosed herein.

Methods, enzymes, recombinant microorganism, and microbial systems are also provided for converting polysaccharides, such as those derived from biomass, into suitable monosaccharides or oligosaccharides, as well as for converting suitable monosaccharides or oligosaccharides into commodity chemicals, such as biofuels. Commodity chemicals produced by the methods described herein are also provided.

    Disclosed herein are methods, systems, and compositions for the pretreatment of biomass within seconds with low inhibitor formation. The pretreatment process is used to convert biomass to a fuel or other useful chemicals by subjecting the feedstock to a rapid retention time under pressure and temperature and/or chemical reactant. The system includes a continuously-operating valve discharge apparatus to discharge pretreated feedstock while maintaining uniform pressure on the pretreatment system.

    The present disclosure provides methods of producing commodity products, the methods involving culturing a host cell that is genetically modified to produce a uronate dehydrogenase (UDH) that converts a sugar acid to its corresponding 1,5-aldonolactone, that uses NADP+ or NAD+ as a cofactor, and that produces NADPH or NADH, respectively, where the host cell coexpresses an endogenous or a heterologous reductase that utilizes the produced NADPH or NADH to generate the commodity product or a precursor thereof. The present disclosure provides a method of producing downstream products of glycerol and pyruvate in a genetically modified microbial host cell, the method involving culturing a genetically modified microbial host cell of the present disclosure in a culture medium comprising D-galacturonic acid. The present disclosure provides variant UDH polypeptides that utilize NADP+, nucleic acids encoding the variant UDH polypeptides; and host cells genetically modified with the nucleic acids.
