核苷酸由碱基、戊糖和磷酸组成,构成生物大分子核酸的基本单位。根据戊糖的不同,核苷酸可分为核糖核苷酸和脱氧核糖核苷酸,进而形成了两种类型的核酸:核糖核酸(RNA)与脱氧核糖核酸(DNA)。核苷酸中的碱基分为嘧啶和嘌呤两类。嘧啶是一种含有两个氮原子的六员杂环有机化合物,常见的有胞嘧啶(C)、尿嘧啶(U)和胸腺嘧啶(T)。而嘌呤则是一种含四个氮的杂环化合物,主要由腺嘌呤(A)和鸟嘌呤(G)构成。核苷酸是在戊糖与碱基缩合形成核苷后,核苷中的糖羟基被磷酸酯化形成的。生物体中游离存在的多为5′-核苷酸,即戊糖的C5羟基被磷酸酯化。
核苷酸通常呈现无色粉末或结晶状,易溶于水而不溶于有机溶剂,在260nm波长下具有强烈的紫外吸收,并且是两性电解质。在不同pH值下解离情况各异,这些特性在核苷酸的制备、分析及测定中具有重要价值。
核苷酸类物质应用广泛,例如ATP和GTP以及5′-核苷酸常作为医疗辅助药物使用;5′-氟尿嘧啶等核苷酸类似物具备抗癌作用;多聚胞苷酸则可诱导干扰素产生,从而发挥抗病毒效果。此外,在食品工业中肌苷酸与鸟苷酸用作味精的增鲜剂,能显著提升风味;而在生物学研究领域,这些物质同样是不可或缺的重要试剂。
