    We report the synthesis and biological activity of a library of aminoalcohol-derived macrocycles from which robotnikinin (17), a binder to and inhibitor of Sonic Hedgehog, was derived. Using an asymmetric alkylation to set a key stereocenter and an RCM reaction to close the macrocycle, we were able to synthesize compounds for testing. High-throughput screening via small-molecule microarray (SMM) technology led to the discovery of a compound capable of binding ShhN. Follow-up chemistry led to a library of macrocycles with enhanced biological activity relative to the original hit compounds. Differences in ring size and stereochemistry, leading to alterations in the mode of binding, may account for differences in the degree of biological activity. These compounds are the first ones reported that inhibit Shh signaling at the ShhN level. (C) 2009 Elsevier Ltd. All rights reserved.

The present invention further provides kits for implementing the above method and the use of an antagonist of a Hedge-hog protein, an antagonist of Smoothened, or an antagonist of Gli for treating the above disease or disorder.

    The present invention relates to a method for culturing epidermis-derived stem cells comprising the step of culturing epidermis-derived stem cells in the presence of a three-dimensional extracellular matrix (3D-ECM) and a basal cell culture medium comprising: Epidermal Growth Factor (EGF); and/or a Vascular Endothelial Growth Factor (VEGF); and/or a Fibroblast Growth Factor (FGF); and further a ROCK (Rho-kinase) inhibitor. The present invention further relates to a method for ex vivo de novo generation of epidermis-derived stem cells. Furthermore, the present invention relates to an epidermis-derived stem cell that is obtainable by a method according to the present invention. Uses of said epidermis-derived stem cell, e.g. uses of said epidermis-derived stem cell for in vitro tissue production, in vitro drug discovery screening and medical applications, are also provided herein. The present invention further relates to a cell culture medium that is employed in the context of a method of the present invention.
