PdII‐catalyzed decarboxylative Heck coupling of α‐alkoxyacrylates with (hetero)aryl carboxylic acids for the stereocontrolled production of (Z)‐β‐heteroarylated vinyl ethers is reported. This methodology offers a rational and step‐economical route to the family of attractive β‐arylated α‐alkoxy α,β‐unsaturated carboxylates, which is a relevant class of building blocks with different applications.

A series of substituted phenylpropanoic acid derivatives was prepared as part of a search for subtype-selective human peroxisome proliferator-activated receptor (PPAR) activators. Structure-activity relationship studies indicated that the substituent at the alpha -position of the carboxyl group plays a key role in determining the potency and the selectivity for PPAR trans activation. The 5-benzylthiazolidine-2,4-dione moiety of insulin sensitising antidiabetic agents can be replaced by a range of alpha-heteroatom functionalised beta-phenylpropanoic acids. alpha-Oxy-carboxylic acids show potent antidiabetic activity and one compound, the alpha-ethoxyacid 15 (SE 213068), is one of the mast potent antihyperglycaemic agents yet reported.
