  A series of chiral heterocyclic aminopyrrolidine derivatives was synthesized as novel melatoninergic ligands. Binding affinity assays were performed on cloned human MT1 and MT2 receptors, stably expressed in NIH3T3 cells. Compound 16 was identified as an orally bioavailable agonist at MT1 and MT2 melatonin receptors with low vasoconstrictive activity. (C) 2003 Elsevier Ltd. All rights reserved.

  本发明公开了一种他司美琼关键中间体(1R,2R)-2-(2,3-二氢苯并呋喃-4-基)环丙甲醇的合成方法，包括步骤：1)1,3-二甲氧基苯、四甲基乙二胺和溶剂，碱作用下加入环氧乙烷反应，得化合物1；2)化合物1酸化关环得化合物2；3)化合物2溶于有机溶剂，碱作用下加入三氟甲磺酸酐，得化合物3；4)丙烯酸乙酯或丙烯酸甲酯和化合物3溶于有机溶剂，碱试剂、钯催化剂作用下和配体反应后得化合物4；5)化合物4溶于有机溶剂，加入还原试剂得化合物5；6)化合物5溶于有机溶剂，加入乙基锌、二碘甲烷和手性配体，通过不对称Simmons–Smith反应即得。该方法能减少合成步骤，提高合成线性总收率，降低成本。

Butyramides 26, 39, 42, 52, 57, and 58 all demonstrated subnanomolar MT2 binding affinity and MT2 selectivity of at least 70-fold over the MT1 receptor.
