Silver(I)-Catalyzed Cascade: Direct Access to Furans from Alkynyloxiranes

Functionalized furans are conveniently formed by a new silver(I)-catalyzed reaction of alk-1-ynyl oxiranes in the presence of p-toluenesulfonic acid and methanol. Evidence supported a cascade mechanism.

Various 2-alkynylallylalcohols were synthesized by a generally applicable Sonogashira coupling protocol. Subsequent gold-catalyzed transformation was investigated. A general approach to 4-methyl-2-substituted-furans is described in which 4-methyl-2-furyl-lithium is the key intermediate. Using this method, pleraplysillin-2, a sesquiterpenoid ester from the sponge Pleraplysilla spinifera, has been synthesised. An alternative, less efficient, route to this type of furan, via acyclic keto-epoxides, is also discussed.

A wide variety of 3-alkyne-1,2-diols have been found to undergo exceptionally clean 5-endo-dig cyclisations followed by dehydration at ambient temperature to give the corresponding furans in essentially quantitative yields when exposed to 10 mol % of 10%,w/w silver(1) nitrate absorbed on silica gel.
