    Ethylacetoacetate enamines of phenylalanine and phenylglycine enhanced the rectal absorption of macromolecular drugs such as lysozyme and heparin in rabbits. Glycerine-1, 3-diacetoacetate, 1, 2-isopropylideneglyceryl-3-acetoacetate and ethylacetoacetylglycolate also enhanced the rectal absorption of lysozyme and heparin. Lysozyme is a basic compound and heparin is an acidic compound, so the non-surfactant adjuvants used in this study can promote the rectal absorption of macromolecular compounds irrespective of their electrochemical properties. The coadministration of calcium ion inhibited the adjuvant action, so complex formation of the adjuvant with calcium ion may occur, reducing the efficacy of these compounds as adjuvants.

    The invention relates to a compound for use in a method of treatment or prevention of migraine and/or symptoms thereof. The compound is selected from beta-hydroxybutyric acid (βHB) or a pharmaceutically acceptable salt thereof, acetoacetate (AcAc) or a pharmaceutically acceptable salt thereof, a metabolic precursor of βHB or AcAc 1,3-butanedio and a compound comprising an acetoacetyl- or 3-hydroxybutyrate moiety. The invention further relates to a pharmaceutical composition comprising βHB, AcAc or a pharmaceutically acceptable salt thereof.

    Transesterification and transthiolesterification of beta-keto esters with variety of alcohols and thiols and selective protection of carbonyl functions with various protecting groups catalyzed by natural kaolinitic clay are described. The clay has been found to be an efficient catalyst in transesterifying long chain alcohols, unsaturated alcohols, and phenols to give their corresponding beta-keto esters in high yields. For the first time, transthiolesterification of beta-keto esters with a variety of thiols has been achieved under catalytic conditions. Clay also catalyzes selective transesterification of beta-keto esters by primary alcohols in the presence of secondary and tertiary alcohols giving corresponding beta-keto esters. A systematic study involving the reactivity of different nucleophiles (alcohols, amines, and thiols) toward beta-keto esters is also described. Sterically hindered carbonyl groups as well as alpha,beta-unsaturated carbonyl groups underwent protection without the deconjugation of the double bond. Chemoselective protection of aldehydes in the presence of ketones has also been achieved over natural kaolinitic clay.
