2-氨基-3-(4-氯苯甲酰基)-4,5-二甲基噻吩可用作有机合成中间体和医药中间体,主要用于实验室研发过程及化工生产过程中。
制备在室温(25℃)下,将0.56 g(17 mmol)硫加入到含有3 g(17 mmol)4-氯苯甲酰基乙腈、3.7 mL(34 mmol)2-丁酮及5.9 mL(68 mmol)吗啉的30 mL乙醇溶液中。将混合物加热至70℃,反应3.5小时后冷却至室温。然后倒入100 mL盐水中混匀,并用乙酸乙酯萃取三次(每次50 mL),合并有机层并用50 mL饱和食盐水洗涤一次。收集有机层,使用无水硫酸钠干燥,过滤后在减压下蒸除溶剂。将残余物进行柱层析,用石油醚-乙酸乙酯(20∶1)洗脱,得到4.3 g黄色固体,收率为92.2%,纯度经HPLC检测为99.5%。熔点:434~436℃;质谱分析:m/z 266.2[M+H]+,288.2[M+Na]+。
