A 5,5‐d2‐luciferin was prepared to measure isotope effects on reactions of two intermediates in firefly bioluminescence: emission by oxyluciferin and elimination of a putative luciferyl adenylate hydroperoxide to dehydroluciferin. A negligible isotopeeffect on bioluminescence provides further support for the belief that the emitting species is the keto‐phenolate of oxyluciferin and rules out its excited‐state

The present invention relates to single and dual reporter luminescence assays utilizing reagents to quench an optical, e.g., an enzyme-mediated luminescence, reaction. In one embodiment of the invention, a reagent is added to an assay which selectively quenches a first enzyme-mediated luminescence reaction without affecting a subsequent distinct enzyme-mediated luminescent reaction(s). An assay kit containing one or more selective quench reagents, and compositions comprising the quench reagent(s), are also provided.

Described are luciferin analogues, methods for making the analogues, kits including the analogues, and methods of using the compounds for the detection of luminescence in luciferase-based assays.
An enhanced luminescent or luminometric assay, especially immunoassay, having a luminescent reaction between a peroxidase enzyme, an oxidant, a 6-hydroxybenzothiazole and a chemiluminescent 2,3-dihydro-1,4-phthalazinedione. The preferred materials are horse radish peroxidase as the enzyme, hydrogen peroxide as the oxidant, 6-hydroxybenzothiazole, 2-cyano-6-hydroxybenzothiazole or firefly luciferin as the benzothiazole and either luminol or isoluminol as the phthalazinedione. In a particularly preferred embodiment the horse radish peroxidase is coupled to an antibody to the substance to be assayed and thereby acts as the assay label in an immunoassay.
