Malaria control programs continue to be threatened by drug resistance. To identify newantimalarials, we conducted a phenotypic screen and identified a novel tetrazole-based series that shows fast-kill kinetics and a relatively low propensity to develop high-level resistance. Preliminary structure–activity relationships were established including identification of a subseries of related amides with
Abstract A key intermediate was obtained as solid through filtration of the reaction mixture of saccharin, chloro(triphenyl)phosphonium chloride, and N,N-diisopropylethylamine (DIPEA) in chloroform. The soluble triphenylphosphine oxide went to filtrate as waste, while the solid was reacted with amines to afford N-sulfonylamidines. In total, 26 N-sulfonylamidine products were obtained in moderate to

Under mild conditions, an N-phthalimidoyl sulfoximine undergoes aminolysis to provide a wide range of 2-formyl benzamides in good to high yields. An unprecedented variant of McFadyen-Stevensreaction explains the product formation. The applicability of the products as building blocks for nitriles and alkynes has been demonstrated.
