    Methods for producing a high resolution image of a sample are provided. In some embodiments, the method includes: detecting first and second sets of spatially-dependent emission signals from a sample labeled with a fluorescent polymeric dye; and producing a high resolution fluorescence image of the sample from the detected first and second sets of spatially-dependent emission signals. In some embodiments, the sample is a cell. Also provided are systems for imaging a sample that include a high resolution light microscope including a light source configured to irradiate a field of view with an excitation light; a photodetector configured to detect an emission signal: and a polarization modulator disposed in the light pathway between the light source and the photodetector; and a sample labelled with a polymeric dye and disposed in the field of view.

    Artificial polynucleotides may have different characteristics than natural polynucleotides so conventional base-calling algorithms may make incorrect base calls. However, because artificial polynucleotides are typically designed to have certain characteristics, the known characteristics of the artificial polynucleotide can be used to modify the base-calling algorithm. This disclosure describes polynucleotide sequencers adapted to sequence artificial polynucleotides by modifying a base-calling algorithm of the polynucleotide sequencer according to known characteristics of the artificial polynucleotides. The base-calling algorithm analyzes raw data generated by a polynucleotide sequencer and identifies which nucleotide base occupies a given position on a polynucleotide strand.

    Methods are provided for detecting a glycosylated target protein in a sample. Aspects of the methods include: (a) contacting a sample comprising a probe-labeled glycosylated target protein with: (i) a first conjugate comprising a first nucleic acid tag linked to a first capture agent that specifically binds the target protein; (ii) a second conjugate comprising a second nucleic acid tag linked to a second capture agent that specifically binds the probe; and (iii) a bridging nucleic acid that hybridizes to the first and second nucleic acid tags; under conditions sufficient to specifically bind the first and second capture agents to the probe-labeled target protein and to hybridize the bridging nucleic acid to the first and second nucleic acid tags to produce a nucleic acid complex; and (b) detecting the nucleic acid complex. Also provided are compositions and kits useful in practicing various embodiments of the subject methods.
