New dihydropyridine derivatives: anti-inflammatory, analgesic and docking studies

The present article describes synthesis of new diethyl 2,6-dimethyl-4-(4-(2-substituted amino-2-oxoethoxy) phenyl)-1,4-dihydropyridine-3,5-dicarboxylates (6a–10b) following multistep synthetic route. Structures of newly synthesized intermediates and title compounds were established by spectral and elemental analyses. 本文介绍了多步合成新的2,6-二甲基-4-(4-(4-(2-取代的氨基-2-氧乙氧基)苯基)-1,4-二氢吡啶-3,5-二羧酸二乙酯(6a – 10b)的方法合成路线。通过光谱和元素分析建立了新合成的中间体和标题化合物的结构。分别通过角叉菜胶诱发的爪水肿和尾巴浸没法筛选最终化合物的体内抗炎和镇痛活性。此外,对活性化合物6c,6d,7d,8和10b进行了分子对接研究为了研究其作用方式,同时体内结果表明,与标准药物相比,这些化合物具有快速起效的抗炎作用并具有突出的活性。带有酰胺官能团的化合物6d和7d显示出最高的抗炎和止痛活性。分子对接的结果强调了体内数据,发现所有对接的分子在纳摩尔级显示出非常低的结合常数。
