Conjugation of ligands to DNA oligonucleotides has been achieved in the solidphase by strain-promoted azide-alkyne cycloaddition (SPAAC). The oligonucleotide, modified with a simple nonfluroinated, monocyclic octyne, efficiently forms conjugates with a range of azide dipoles with varying steric and electronic characteristics.
