    Phthalates are used primarily as plasticizers to make polyvinyl chloride (PVC) soft and flexible. In recent years the phthalate esters have attracted increasing attention as environmental and biomedical pollutants and, because of their toxicological characteristics. In particular, they are more and more recognized as endocrine disrupters. In this context, we describe herein an efficient synthetic pathway leading to a series of metabolites of di(2-ethylhexyl) phthalate (DEHP). Mono(2-ethylhexenyl) phthalate was used as starting material to obtain these products in good yield, large scale and GC-MS purity. The metabolites of DEHP were synthesized, for the first time, as biomarkers to verify their quantitative determination in human urine and serum by GC-MS analysis for studying the exposure to phthalates and establishing reference values.     Two ring-deuterated major metabolites of bis(2-ethylhexyl) phthalate were obtained in excellent yields by means of an improved oxidation procedure. The by-product of their interconversion by Swern oxidation is elucidated and a possible mechanism of its formation is proposed.

    The synthesis of two oxidized metabolites of bis-(2-ethylhexyl)-phthalate is described. The target structures were obtained by esterification of the appropriate alcohol carrying a protected hydroxy group with phthalic anhydride, followed by deprotection and further oxidation.

    A method of assessing the risk in female patients of having neurocognitive developmentally disabled children. The method including measuring the toxin concentrations in the urine of a female subject wishing to become pregnant, or of a subject that is pregnant. Quantify the risk of neurocognitive developmentally disabled children born to the subject by comparing the toxin concentrations in the subject to reference concentrations utilizing a weighting algorithm. Advise the patient of the quantified risk of having neurocognitive developmentally disabled children. The concentrations of dialkyl phosphates, endocrine disruptors, and xenoestrogens compounds are evaluated. Additionally, the level of PON1 activity may be included in the risk assessment weighting algorithm.

    The present invention relates to methods and systems for assessing ovarian reserve and function in a female subject and informing course of treatment thereof. The invention provides methods for assessing ovarian reserve and function by analyzing both clinical and genetic data/characteristics from a female subject. These methods involve the determination of the presence of one or more mutations in a gene, the gene being associated with fertility and/or ovarian reserve or function. In certain aspects the methods also involve the determination of one or more clinical characteristics associated with fertility and/or ovarian reserve or function. In certain embodiments, the clinical and genetic characteristics obtained from a female subject can be used as data to be input to an ovarian reserve predictor, such that a probability of the female subject suffering from ovarian reserve dysfunction or premature decline can be generated.
