    摘要:
    Microcystin LR metabolism in mammals was examined. The degradation of microcystin LR by the enzymes, pepsin, trypsin and chymotrypsin, from porcine or human gastrointestinal tract was examined. Microcystin LR was digested by these proteases, but 3-amino-2, 6, 8-trimethyl-10-phenyldeca-4E, 6E-dienoic acid (DmADDA) was not formed from microcystin LR treated with pepsin and chymotrypsin. The formation of DmADDA was detected from microcystin LR treated with trypsin. Furthermore, DmADDA was detected from the male ddY mice liver orally administered microcystin LR.     Methods for detecting and quantifying one or more microcystin compounds in a sample are described. The methods may include a preconcentration step, and generally utilize an LC-MS or LC-MS/MS analysis with an Orbitrap Fusion mass spectrometer. The methods provide excellent recoveries and limits of quantification of microcystins.

    A method is used for producing a modified non-ribosomal peptide, e.g. a modified microcystin and/or modified nodularin (together CA), including the steps of: a) growing a modified non-ribosomal peptide producing cyanobacteria strain in a culture media, h) adding one or more modified substrates, preferably modified amino acids to said culture, and c) inoculating the non-ribosomal peptide, producing strain the presence of said modified substrates. The thus modified non-ribosomal peptide may be used for the therapy of various diseases.
