Synthesis and in Vitro Transdermal Penetration Enhancing Activity of Lactam N-Acetic Acid Esters

A homologous series of N-acetic acid esters of 2-pyrrolidinone and 2-piperidinone has been prepared and evaluated for its ability to enhance the skin content and flux of hydrocortisone 21-acetate in hairless mouse skin in vitro. Enhancement ratios (ER) were determined for flux (J) 24-hour diffusion cell receptor cell concentrations (Q(24)), and 24-h full-thickness mouse skin steroid content (SC) and compared to control values (no enhancer present). In addition, in an attempt to abrogate toxicity, these dermal penetration enhancers were designed to have the potential for biodegradation by dermal esterases. 2-Oxopyrrolidine-alpha-acetic acid dodecyl ester (5) showed the highest enhancement ratios for J(ER 67.33) and Q(24) (ER 180.66). 2-Oxopipelidine-alpha-acetic acid decyl ester (10) showed a high Q(24) (ER 162.07) but a lower J (ER 12.67). 2-Oxopyrrolidine-alpha-acetic acid decyl ester (3) showed the highest enhancement ratio for SC (ER 8.7). The ER Q(24) for 3, 5 and 10, as well as other lactam N-acetic acid esters in this work, were significantly higher than the ER found using Atone as enhancer.(dagger)
Photostable UV-photoprotecting cosmetic sunscreen compositions contain (a) an effective UV-photoprotecting amount of at least one dibenzoylmethane compound sunscreen, (b) an effective radiation-photostablizing amount of at least one pyrrolidinone compound having the formula (I) below:
and, advantageously, at least one liquid fatty phase and at least one lipophilic active sunscreen agent of low solubility, formulated into (c) a cosmetically acceptable support therefor.

La présente invention est relative à une composition cosmétique contenant l'association d'au moins un dérivé du dibenzoylméthane et d'au moins (b) un dérivé de pyrrolidinone de formule (I) suivante :

R1 désigne un radical aryle en C6-C20 éventuellement substitué par une chaîne alkyle linéaire ou ramifié en C1-C20 ; un radical alkyle en C1-C20, linéaire ou ramifié,
n = 0 ou 1 ; sous réserve que :
- lorsque n = 1, alors le radical R1 ne peut pas être un radical aryle,
- lorsque n = 0, alors R1 ne peut pas être un radical alkyle en C1-C2
Elle concerne également un procédé de photostabilisation vis-à-vis du rayonnement d'au moins un dérivé du dibenzoylméthane par une quantité efficace d'au moins un dérivé de pyrrolidinone de formule (I).
La présente invention est relative également à l'utilisation d'au moins un dérivé de pyrrolidinone de formule (I), dans une composition comprenant dans un milieu cosmétiquement acceptable au moins une phase grasse liquide et au moins un actif lipophile de faible solubilité et en particulier un filtre organique UV de faible solubilité, comme solvant dudit actif dans ladite phase grasse liquide.
