    Plug-and-Play Fluorophores Extend the Spectral Properties of Spinach

    Spinach and Spinach2 are RNA aptamers that can be used for the genetic encoding of fluorescent RNA. Spinach2 binds and activates the fluorescence of (Z)-4-(3,5-difluoro-4-hydroxybenzylidene)-1,2-dimethyl-1H-imidazol-5 (4H)-one (DFHBI), allowing the dynamic localizations of Spinach2-tagged RNAs to be imaged in live cells. The spectral properties of Spinach2 are limited by DFHBI, which produces fluorescence that is bluish-green and is not optimized for filters commonly used in fluorescence microscopes. Here we characterize the structural features that are required for fluorophore binding to Spinach2 and describe novel fluorophores that bind and are switched to a fluorescent state by Spinach2. These diverse Spinach2 fluorophore complexes exhibit fluorescence that is more compatible with existing microscopy filter sets and allows Spinach2-tagged constructs to be imaged with either GFP or YFP filter cubes. Thus, these "plug-and-play" fluorophores allow the spectral properties of Spinach2 to be altered on the basis of the specific spectral needs of the experiment.

    The present invention relates to a novel probe set for an isothermal one-pot reaction, and uses thereof and, particularly, provides a method for easily, accurately, and quickly diagnosing molecules, in particular, infection diseases, in the field, the method having a form applicable in the field on the basis of a nucleic acid sequence by using a one-pot reaction composition under an isothermal condition. If a molecular diagnostic platform according to the present invention is used, a target sequence can be quickly and accurately detected. Also, elements (reaction buffer, enzyme) needed in a diagnostic process using the present invention are much more simple and convenient than in a conventional antibody-based diagnosis, and, thus, the diagnostic process can be performed by a non-skilled person, and the sensitivity and speed of diagnosis can be increased, as all reactions are unified at a constant temperature without expensive equipment used in general nucleic-acid-based molecular diagnostic technology, and an amplification process is performed automatically during a reaction process.

    Provided herein are imaging probes and systems and methods employing such imaging probes for real-time, label-free, multiplexed imaging of RNAs in living cells. More particularly, aptamer-based sensors (“aptasensors”) and molecular fuses comprising multiple aptasensors are genetically encoded imaging probes comprising RNA-target binding sequence and an intramolecular reconfiguration sequence. The probe is configured such that binding of a RNA target by the RNA-target binding sequence triggers the intramolecular reconfiguration sequence to reconfigure such that an optically detectable output is generated by the probe.
