    The present invention relates to a photolabile compound including a coumarin family caging group covalently bound to an organic moiety containing a carboxyl group, an amino group, a sulfhydryl group, and/or a hydroxyl group. The photolabile compound, upon absorbing visible light, releases the organic moiety. Also disclosed is a method of making a photolabile compound that releases a biologically active compound upon absorbing visible light by providing an organic moiety and binding a coumarin family caging group to the organic moiety under conditions effective to make the photolabile compound. The present invention also relates to a method of obtaining a biologically active compound in a sample. The method involves adding to a sample a photolabile compound including a coumarin family caging group covalently bound to an organic moiety. The sample is then illuminated with visible light under conditions effective to obtain a biologically active compound including the organic moiety.

    Methods, devices, and systems for measuring the concentration of glycated hemoglobin (HbA 1 c) in a blood sample are provided. A blood sample may be contacted with a lysing agent, an assay to measure total Hb, and an assay to measure HbA 1 c, which comprises donor and acceptor dyes attached to a Hb specific antibody and a HbA 1 c analog. Preferably, the emission spectrum of the donor dye overlaps with the excitation spectrum of the acceptor dye. The concentration of HbA 1 c in the sample may be correlated with an increase in donor dye and/or a decrease in acceptor dye fluorescence in the presence of HbA 1 c as compared to the donor and/or the acceptor dye fluorescence in the absence of HbA 1 c.

    The invention provides transdermal optical analysis systems, test sensors, methods, and kits for determining the presence and/or concentration of at least one analyte in a fluid sample. The system includes a transdermal test sensor including an aqueous material including at least one analyte selective reagent and at least one optically active moiety. The optical system preferably uses fluorescent spectroscopy to correlate fluorescent emission or adsorption from a dye with the analyte concentration of the sample. An optical light source and/or detector may be housed with the aqueous material in a housing or external to the housing of the aqueous material.

    A transdermal test sensor includes a test chamber including a liquid, a reagent system in contact with the liquid, a housing containing the liquid, and a semipermeable membrane. The housing includes an opening, the semipermeable membrane is connected to the housing and covers the opening, and the housing and the semipermeable membrane enclose the liquid and the reagent system. The transdermal test sensor also includes an analyzer in communication with the liquid. When porated tissue is contacted with the semipermeable membrane and sufficient time is allowed for a fluid sample to traverse the porated tissue and for an analyte in the fluid sample to enter the liquid in the transdermal sensor through the semipermeable membrane, a change in at least one optical property or at least one electrical property of the liquid is detected. The change detected is then correlated with the analyte concentration in the fluid sample.
