New green fluorescent protein (GFP) chromophore analogues, namely 4-(diarylmethylene)imidazolinones (DAINs), were readily synthesized under weakly acidic conditions using a novel condensation reaction between methyl imidate (or thioimidate) and ethyl N-(diarylmethylene)glycinate.
