‐Selective Diels–Alder Route to Hedgehog Pathway Inhibitors
The first synthetic access to stable and acyclic 1,3‐butadienes with two electron‐withdrawing carbonylgroups and their potential to deliver new molecular scaffolds through intriguing endo‐selective Diels–Alder cycloadditions are presented. The bicyclic scaffolds produced through the cycloaddition chemistry of electron‐deficient dienes afforded potent Hedgehog signaling pathway inhibitors.

The first report on electrochemical AuI/AuIII catalysis for 1,2-difunctionalization of C−C multiplebonds has been presented. This external-oxidant-free approach utilizes the anodic oxidation of vinyl-AuI to vinyl-AuIII complexes to achieve oxy-alkynylation of allenoates to access alkyne-substituted butenolides in an undivided cell.
