An optical sensor (10) is used to determine the presence and/or amount of a target in a sample liquid (20). The optical sensor (10) has an optical sensing layer (12) including, in the same layer, a fluorescent reporter (36), an optical isolating agent (40), and optionally a fluorescent reference (38). The optical sensing layer (12) has an outer surface (26) that contacts the sample liquid in a detection chamber (14) and an opposing surface through which a light source (28) irradiates the optical sensing layer (12) with excitation light (30) and a detector (32) detects fluorescence (34) emitted from within the optical sensing layer (12). The optical isolating agent (40) reduces the amount of (i) excitation light (30) that passes through the optical sensing layer (12) and reaches the sample liquid and (ii) background fluorescence that is emitted within the sample liquid and passes back through the optical sensing layer (12) to the detector (32). Accordingly, the optical reporter fluorescence (36), and optionally optical reference fluorescence, can be detected with higher signal to noise ratios than in the absence of the optical isolating agent (40).
