  Aureothin 是来自 Streptomyces thioluteus 的莽草酸-聚酮化合物杂化代谢物，具有稀有的硝基芳基部分、手性四氢呋喃环和 O-甲基化吡喃酮环。金黄色素的抗菌和抗肿瘤活性引起了我们对调节其结构和生物活性特征的兴趣。在使用突变合成、生物转化和组合生物合成的综合方法中，生成了定义的金黄素类似物库。利用聚酮合酶组装线对不同起始单元的混杂性以及吡喃酮和四氢呋喃环形成的可塑性。在制备规模上生产了 15 种在芳基残基、吡喃酮环和氧化主链上进行了修饰的新金黄素类似物，并对其进行了充分表征。值得注意的是，各种新的金红素衍生物的细胞毒性比金红素小，但具有更好的抗增殖活性。此外，我们发现 THF 环对于金霉素类似物对某些病原真菌的显着选择性活性至关重要。
