Cycloaddition reactions of cycloheptatrienone and ten dienes were examined to define diene substituent effects in the thermal [6+4] cycloaddition reaction of cycloheptatrienone. The [6+4] adducts from dienes with greater electron density were formed more rapidly and were thermodynamically less stable than the adducts from more electron-poor dienes. A thermally allowed tropone-diene [6pi + 4pi] cycloaddition is employed to produce a functionally rich BC ring building block for the construction of 8-isoingenol intermediates. Bridgehead enolate alkylation followed by aldol condensation is used to install the elements of the A-ring. Stereo- and regioselective bis-hydroxylation at C-13 and -14 of the C-ring followed by activation as the corresponding bis-mesyloxy derivative and treatment with diethyl sodiomalonate affords an advanced, functionalized isoingenol tetracyclic intermediate.
