  摘要：

  MG-132是三肽醛（Z- 1-亮氨酸-1-亮氨酸-1-亮氨酸-H，2）蛋白酶体抑制剂，具有抗肿瘤活性并增强化学疗法和放射疗法的细胞生长抑制作用/细胞毒性作用。由于具有非天然构型和氨基酸修饰的侧链的三肽合成麻烦，因此仅报道了MG-132的两种立体异构体。在这里，我们提出了一种基于Ugi反应合成三肽醛的新方法。手性，对映体稳定的乙酸2-异氰基-4-甲基戊酯被用作Ugi反应的底物，导致三肽骨架的形成。所得产物的进一步官能化导致三肽醛的合成。合成了MG-132的所有立体异构体，并研究了它们作为胰凝乳蛋白酶样，胰蛋白酶样和肽基谷氨酰胺肽水解蛋白酶活性的潜在抑制剂。S，R，S）-（-）- 2立体异构体是比MG-132更有效的蛋白酶体抑制剂。
