We describe a new reduction process for the efficient preparation of various, especially 2-substituted, piperidin-4-ones which are not accessible by other reaction pathways until now. Tetrahydropyridinylidene salts (THPS) with differing substitution pattern are the educts of this reaction. These compounds are available by mainly two different reaction sequences and they can be further modified by insertion
The present invention relates to a process for preparing a pi-peridin-4-one comprising a step of reducing a tetrahydropyridin-4-ylidene ammonium salt of formula (I)
to a compound of formula (II)
as well as a compound according to formula (II) and a method of derivatizing a piperidin-4-one.

Synthesis of 2-substituted bamipine derivatives

2-Substituted derivatives of bamipine and its 1-phenyl analogue have been prepared in several steps from dihydropyridine-2(1H)-thiones. The configurations and the conformations of the formed diastereoisomers have been investigated by NMR spectroscopy. The antimycobacterial activities of the title compounds have been examined.
1-Unsubstituted 4-dimethylamino-5,6-dihydropyridine-2(1H)-thiones were converted to isomeric piperidin-4-ols which were separated and N-methylated to 2-substituted 1-methylpiperidin-4-ols. Their 1-phenyl analogues were prepared from 4-dimethylamino-5,6-dihydro-1-phenylpyridine-2(1H)-thiones. After their conversion to dihydro-1-phenylpyridin-4(1H)-ones the hydrogenation gave isomeric 1-phenylpiperidin-4-ols
