    Phenylalanine 93 of the human UGT1A10 plays a major role in the interactions of the enzyme with estrogens

    Little is currently known about the substrate binding site of the human UDP-glucuronosyltransferases (UGTs) and the structural elements that affect their complex substrate selectivity. In order to further understand and extend our earlier findings with phenylalanines 90 and 93 of UGT1A10, we have replaced each of them with Gly, Ala, Val, Leu, Ile or Tyr, and tested the activity of the resulting 12 mutants toward eight different substrates. Apart from scopoletin glucuronidation, the F90 mutants other than F90L were nearly inactive, while the F93 mutants' activity was strongly substrate dependent. Hence, F93L displayed high entacapone and 1-naphthol glucuronidation rates, whereas F93G, which was nearly inactive in entacapone glucuronidation, was highly active toward estradiol, estriol and even ethinylestradiol, a synthetic estrogen that is a poor substrate for the wild-type UGT1A10. Kinetic analyses of 4-nitrophenol, estradiol and ethinylestradiol glucuronidation by the mutants that catalyzed the respective reactions at considerable rates, revealed increased K-m, values for 4-nitrophenol and estradiol in all the mutants, whilst the K-m values of F93G and F93A for ethinylestradiol were lower than in control UGT1A10. Based on the activity results and a new molecular model of UGT1A10, it is suggested that both F90 and F93 are located in a surface helix at the far end of the substrate binding site. Nevertheless, only F93 directly affects the selectivity of UCT1A10 toward large and rigid estrogens, particularly those with substitutions at the D ring. The effects of F93 mutations on the glucuronidation of smaller or less rigid substrates are indirect, however. (C) 2011 Elsevier Inc. All rights reserved.

    The present invention is directed to a homogeneous fluorescence polarization immunoassay for determining total estriol content in body fluids, to the various components needed for preparing and carrying out such an assay, and to methods of making these components. Specifically, tracers, immunogens and antibodies are disclosed, as well as methods for preparing them. The assay is conducted by measuring the degree of polarization of plane polarized light that has been passed through a sample containing antiserum and tracer. The combination of antiserum and tracer used will possess excellent cross-reactivity to estriol-3-sulfate, eliminating the need for enzyme hydrolysis of this conjugate to free estriol.

    Methods, devices and test kits for monitoring the ovulation cycle, involve testing the body fluid, e.g. urinary, concentration of one or more analytes. Preferably estrone-3-glucuronide and luteinizing hormone are both measured, and a reference concentration for E3G is established at about day 6 of the current cycle. Preferably, disposable testing devices are used, in conjunction with a relatively permanent electronic reader/monitor. The number of "daily" tests required per month can be minimised.

    A method of monitoring the status of a current ovulation cycle of an individual female mammalian (generally human) subject, involving repeated testing of the body fluid concentration of at least one analyte, preferably estrone-3-glucuronide (E3G) of significance in relation to the status of the ovulation cycle, during at least the pre-ovulation phase of the current ovulation cycle of the individual subject, wherein testing for said analyte concentration during the current ovulation cycle conducted at least once during the interval spanning days 1 to 7 inclusive following the onset of menses, to establish a reference concentration value for analyte in the current cycle, and then testing is conducted repeatedly during a plurality of days, preferably commencing at least 5 numerical days in advance of the mean numerical day on which actual ovulation has occurred over one or more previous ovulation cycles in the same individual subject, analyte concentration values obtained during said repeated testing being compared with the reference concentration value to determine whether a concentration change indicative of imminent ovulation is occurring or has occurred since the previous test. The method can be based solely on E3G measurements.
