A straightforward and efficient method for the palladium-catalyzed direct cross-coupling of quinones with N,N′-dialkyluracils via 2-fold C–H activation has been developed to rapidly construct uracil substituted quinone structural motifs.

Pd-catalyzed regio- and stereoselective C-nucleoside synthesis from pyranoid glycals and unactivated N,N-dialkyluracils is described. Depending upon the reaction conditions, either β-hydride or β-heteroatom eliminated C-nucleosides were obtained exclusively. Different glycals with various protecting groups reacted smoothly. A novel procedure for the solid-phasesynthesis of 1,3,6-trisubstituted uracils using a polymer-supported selenium resin is described. The advantages of the new method are good yields, high purity, straightforward operations, broad range and high diversity of the products, lack of odor, and good stability of the resins.

The Pd-catalyzed cross-dehydrogenativecoupling (CDC) between two heteroarenes was developed. A variety of benzofuran and uracil derivatives were constructed through C–C bond formation by using CDC reactions. The substrates did not require prefunctionalization, and the products were obtained in good yields with good regioselectivities at roomtemperature.

A highly efficient protocol for the regioselective dehydrogenative cross-coupling of pyridine-N-oxides with N,N-dialkyl uracils via 2-fold C–H activation has been developed. The resultant uracil substituted pyridine-N-oxide structural motifs is readily deoxygenated, making the present procedure a highly attractive route for the synthesis of 5-pyridine substituted uracils.
