The present invention relates to novel compounds of formula (I), and compositions capable of inhibiting PHD1 enzyme activity selectively over other isoforms, for example, PHD2 and/or PHD3 enzymes. The direct asymmetric conjugate addition of α-cyanoacetates to enones generating densely functionalized α-amino acid precursors with adjacent quaternary and tertiary stereocenters is described comparing mono- and bis-palladacycle catalysts. This edge article features the complementary value of mono- and bimetallic catalysis in a case study using related catalyst systems. Different major diastereomers
no]acetates with quaternary stereocenters and potential antibacterial activities were obtained in excellent yields with good to excellent ee values under as low as 0.05 mol % catalyst loading. The products could be easily transformed to useful α‐amino amides and 1,2‐diamines. Besides, a possible transition state model was proposed to elucidate the origin of the chirality induction.
