A dry type multilayer analysis element for assaying a concentration of a specific component utilizing a competitive immune reaction comprises a detection element comprising a detection layer which receives a labelled complex formed as a result of the competitive immune reaction or an optically detectable change formed dependant upon an amount of the labelled complex of the specific component and having further provided thereon the detection layer a reaction layer comprising a fibrous porous medium. The multilayer analysis element absorbs an amount of a sample solution necessary for the competitive immune reaction so that the multilayer analysis element has high sensitivity and high reproducibility.

A multilayer analytical element for non-isotopic assay of an analyte contained in an aqueous liquid sample, utilizing a competitive binding reaction to a protein between the analyte and a non-isotopically labeled analyte substance or analogue thereof, which comprises:
a light-transmissive water-impermeable support;
a sheet provided on the support, which comprises a continuous binder matrix containing the non-isotopically labeled analyte substance or analogue thereof;
a porous reaction sheet provided on the binder matrix sheet, which contains a predetermined amount of the protein fixed therein, the protein being specifically bindable to the analyte; and
a porous sharing sheet provided on the reaction sheet,
in which the substantial separation of the bound non-isotopically labeled complex (B) from the free non-isotopically labeled analyte substance or analogue thereof (F) is accomplished by the presence of said porous sharing sheet.
A slide enclosing the multilayer analytical element and a non-isotopic assay employing the multilayer analytical element are also disclosed.

At the conclusion of a selective binding assay (e.g., immunological or nucleic acid), an enzyme is present in modulated concentration and/or activity to moderate a chemical reaction such as the cleavage of o-nitrophenyl-β-D-galactopyranoside. After a controlled period, the enzymatic product is transferred to the gas phase and concentrated relative to other components in the enzymatic reaction mixture, such as by extraction into ethyl ether, injection into a gas chromatography column and detection by flame ionization or electron capture. Kits for such assays are also disclosed.

Fluorescent labels comprise a polysaccharide bound to a polymeric particle which contains a fluorescent rare earth chelate. These labels can be attached to any of a variety of physiologically reactive species to provide labeled species which have improved stability in aqueous solutions. The labeled species are particularly useful in specific binding assays to determine an immunologically reactive ligand, e.g. a hapten, in either solution or dry analytical techniques.
