    Structural Optimization and Biological Evaluation of Substituted Bisphenol A Derivatives as β-Amyloid Peptide Aggregation Inhibitors

    The aggregation of A beta is a crucial step in the etiology of Alzheimer's disease. Our previous work showed that A beta undergoes alpha-helix/beta-sheet intermediate structures during the conformational transition, and an A beta aggregation inhibitor (1) was discovered by targeting the intermediates. Here, structure optimization toward compound 1 was performed and 34 novel derivatives were designed and synthesized. Nine compounds showed more effective inhibitory activity than the hit compound 1 in ThT fluorescence assay. Among them, compound 43 demonstrated more excellent inhibitory potency, which not only can suppress the aggregation of A beta but also can dissolve the preformed fibrils as shown by CD spectroscopy, PICUP and AFM assays. Cellular assay indicated that 43 has no toxicity to neuronal cells, moreover, can effectively inhibit A beta(1-42)-induced neutrotoxicity and increase the cell viability. Together, on the basis of these positive results, these novel chemical structures may provide a promising potential for therapeutic applications in AD and other types of neurodegenerative disorders.

    The invention is a membrane comprising a dense skin of a polyestercarbonate, wherein the diphenolic residue in the backbone of the polyestercarbonate is a tetrabromo diphenolic residue, wherein the membrane has a separation factor for oxygen and nitrogen of 6.5 or greater at 24°C. In another aspect, the invention is a method of separating oxygen from a mixture of gases comprising nitrogen and oxygen, wherein the method comprises (a) contacting the gas mixture with one side of a thin, non-porous membrane comprising a polyestercarbonate described hereinbefore while maintaining a pressure difference across the membrane with the lower pressure being on the other side of the membrane; and (b) removing the permeated oxygen from the other side of the membrane, wherein separation factor for the oxygen and nitrogen is 6.5 or greater at 24°C. The membranes of this invention demonstrate surprisingly high separation factors for oxygen and nitrogen separations. The membranes of this invention have good mechanical properties and therefore are useful under more extreme conditions, for example temperature and pressure, than many commercially used membranes.

    The present invention provides an electro-optical device comprising NLO polymer in the active layer with improved stability for light in the wavelength range of 600-1600 nm is guided through the device. The present invention is directed to an electro-optical device comprising two cladding layers and an active layer sandwiched between these cladding layers, with the active layer comprising a non-linear optically active side chain polymer with NLO-chromophores having an N-donor-delocated π system-acceptor structure, the N-donor not being bonded directly to alkyl groups. There are two ways to create NLO-chromophores with N-donors which do not have adjacent linear alkyl groups: The first way is to bond one or two aromatic groups. The N-atom is further bonded to the delocated π system. If only one aromatic group is present, the N-atom is, besides the delocated system, further bonded to a hydrogen or deuterium atom. The second way of avoiding adjacent alkyl groups is to employ an N-donor moiety wherein no adjacent linear alkyl groups are present.
