Disclosed is a new resolution process of (S)-3-aminomethyl-5-methylhexanoic acid, which process comprises the following steps: employing benzoyl-L-glutamic acid, p-methylbenzoyl-L-glutamic acid, benzenesulfonyl-L-glutamic acid or p-methylbenzenesulfonyl-L-glutamic acid as resolution agents to resolve racemic 3-aminomethyl-5-methylhexanoic acid for the first time, to give the first resolution product, resolving the first resolution product for the second time by the same resolution agent as that for the first resolution, to give the second resolution product, and thus obtaining a resolution salt-forming product, acidly hydrolyzing the resolution salt-forming product, extracting and separating the resolution agent, adjusting the pH-value to neutral, distilling to give the product (S)-3-aminomethyl-5-methylhexanoic acid. The process has such advantages as little environmental contamination, high effect and steady, easy operation, high product purity, low cost and being suitable for large-scale produce.
