    The interaction of a cobalt porphyrin with cancer-associated nitrosamines

    A cobalt porphyrin (CY-B) was presented, and its interaction with tobacco-specific nitrosamines (TSNAs) was investigated by UV-Vis spectroscopy and high-resolution mass spectrometry. The results revealed that the stoichiometry of the host-guest interaction was 1:2 and that the binding constant between CY-B and TSNAs was within the range of 0.78×10(8)-7.83×10(8)M(-2). The coordination strength between CY-B and TSNAs decreased in the sequence of NNN>NAB>NAT>NNK based on the binding constant. The interaction mechanism of CY-B with TSNAs involved a coordination interaction, and the π-π interaction between the porphyrin macrocycle and the aromatic frame of the TSNAs pyridines may also have been a driving force. The measured thermodynamic properties demonstrated that the reaction of CY-B with TSNAs was spontaneous and that the driving force for the interaction was a change in enthalpy. The reaction was exothermic, and an increasing temperature inhibited the interaction. The IR spectrum of the complex revealed that the NNO group of TSNAs and the metal cobalt of CY-B formed the six-coordinate complex.

    A class of molecularly imprinted polymers that specifically recognizes and binds to TSNAs are useful, for example, in analysis and separation of TSNAs from biological fluids. Such polymers are also useful in methods of treating and manufacturing tobacco products and materials. Also disclosed are selectively adsorbent non-molecularly imprinted polymer of a non-acidic monomer and a cross-linker having polar functionality, one of which is hydrophilic, the other of which is hydrophobic, and the use of such polymers in the treatment of materials containing nicotine and nitrosamine.

    Porous beads are provided of a polymer of a non-acidic monomer and a cross-linker having polar functionality, one of which is hydrophilic, the other of which is hydrophobic, further comprising residues of polyvinyl alcohol. The beads may be molecularly imprinted or non-molecularly imprinted. The use of such beads in the removal of nitroso-containing compounds from material containing them is also disclosed. Also disclosed is a non-molecularly imprinted polymer which is selectively adsorbent for at least one tobacco specific nitrosamine in the presence of nicotine, said polymer being a polymerization product of a non-acidic monomer and a cross-linker having polar functionality, one of which is hydrophilic, the other of which is hydrophobic.

    The invention relates to method of treating a material containing nicotine and at least one nitrosamine to reduce the quantity of nitrosamine therein and to methods of producing a tobacco extract with reduced nitrosamine content. The methods comprise contacting tobacco with a solvent to produce a liquid phase tobacco extract containing nitrosamines and treating the liquid phase tobacco extract to decompose the nitrosamines in the liquid phase. The liquid phase extract may then be treated to reduce nitrosamine decomposition products therein. The invention also relates to tobacco material produced using these methods and to tobacco products incorporating such tobacco material.
