  通过两种方法制备高度官能化的噻吩。第一种方法是在三取代的5-溴噻吩25上进行锂-卤素交换反应，生成相应的5-硫代噻吩26，然后使其与二甲基甲酰胺或甲醛反应，以高收率得到5-甲酰基28或5-羟甲基噻吩衍生物31。将它们进一步转化为其他四取代的噻吩。第二种方法由三个组分组装噻吩环：苄硫醇，醛和乙烯基膦酸酯10。因此，使苄硫醇二锂化，然后与适当的醛反应，得到2-巯基-2-苯基乙醇衍生物37。迈克尔加成37至10，然后氧化羟基，得到酮膦酸酯39。分子内维蒂希型反应产生噻吩骨架。
